N4-Etheno-dC CEP (BA 0391), enables the incorporation of a fluorescent pyrimidine nucleoside analog into oligonucleotides for the investigation of nucleic acid interactions. Due to the similarity in structure between dC and 3, N4-Etheno-dC, the modified nucleoside can be incorporated into DNA with retention of DNA structure and nucleobase recognition.43, N4-Ethenocytosine is reported to be a highly mutagenic DNA adduct that is recognized and excised by the double-stranded uracil DNAglycolase in E. Coli, and by human thymine-DNA glycolase proteins in vitro. The miscoding potential in reactions catalyzed by mammalian DNA polymerases for3, N4-etheno2'-deoxycytidine has also been reported. Both BA 0403and BA 0391 can be efficiently incorporated into oligonucleotides using standard conditions and taking care to avoid prolonged exposure to ammonium hydroxide.
