8-oxo-dG-CE Phosphoramidite allows investigation of the structure and activity of oligonucleotides containing an 8-oxo mutation. This is formed naturally when DNA is subjected to oxidative conditions or ionising radiation. The resulting 8-oxo modification is significant in mutagenesis and ultimately carcinogenesis.

Physical & Dilution Data
Dilution volumes (in ml) are for 0.1M solutions in dry acetonitrile (LK4050). Adjust accordingly for other concentrations. For µmol pack sizes, products should be diluted as 100µmol/ml to achieve 0.1M, regardless of molecular weight.

Coupling
No changes are required from the standard method recommended by the synthesiser manufacturer. Coupling is as per standard nucleoside amidites.
Deprotection
Cleave and deprotect with ammonium hydroxide containing 0.25M 2-mercaptoethanol for 17h at 55°C to avoid oxidative degradation of the 8-oxo-dG site.
Storage & Stability
Store in the fridge, dry. Stability in solution is 24h.
